Toluidine staining of resin or plastic embedded peripheral nerve.
Toluidine blue is a simple metachromatic stain used to visualize endoneurial elements in resin or plastic embedded specimens. Nuclei, myelin, and red blood cells stain dark blue. Cell cytoplasm and extracellular protein stains light blue, and axon cylinders stain very light blue to white. This section of peripheral nerve was obtained from a patient with hereditary sensory and motor neuropathy type I (Charcot-Marie-Tooth disease), and demonstrates concentric layers of Schwann cells around either normal, thinly myelinated, or demyelinated axons.
